Jewelpet Twinkle☆ takes place in Jewel Land, where Jewelpets with the ability to use magic live alongside Witches. The School of Witchcraft and Wizardry is where these creatures learn to harness their magical powers with the aid of Jewel Eyes. Ruby, a white Japanese Hare Jewelpet, has difficulty controlling her magic, which leads to her being sent to the Human World in error after using a special card given by the magicians.
